If a valve is autoclaved between 121-125 deg C, do I need to wait before closing the valve?

No, you can close the valve anytime after autoclaving.

The diaphragm materials used in Asepco valves are rated for temperatures of 135 deg C or more, depending on material.  Additionally, all actuators have built-in non-adjustable travel stops to prevent over-tightening of the diaphragm.
